Injection

1. Pull needles using program #88

2. Spin 10ul of DNA at max for 20′ (less will probably work)

3. Wipe open end of needle with kimwipe

4. Pipette 0.5ul of DNA to open end of needle, wipe needle again

5. Press menu button to change needle

6. Find needle by closing light on scope

7. Break needle on side over another needle in oil (halocarbon oil 700)

8. Press menu again, press clean to inject (should be a bubble)

9. Breathe on 2% agarose on coverslip

10. Pick worms with oil, then dab worms on oil again before placing on slide

11. Worms should be well-fed young adults with one row of eggs. Gonad should be visible (2 clearings on the dorsal side).

12. Inject worms in the middle of the gonad. The clearing should have “stones.” Stones all over the gonad typifies the surface of the gonad. The middle of the gonad should just be one row of stones on the side of the worm.

13. See the turn

14. Recover worm with 0.5ul of M9 and a hook made from flaming the pointed end of a needle.

15. 3 injected worms per plate

16. A couple days later find F1s with co-injection marker and place on individual plates. % very low usually

17. Find F2s that have transmitted. % that transmit is low.
