Lab - Gregarines
1. Gregarina sp. Syzygys of trophozoites. Identify primite, satellite, protomerite, deuteromerite, nucleus, septum. 2. Gregarina sp. Syzygys of trophozoites. Identify primite, satellite, protomerite, deuteromerite, nucleus, septum. 3. Section of midgut of meal worm (Tenebrio) larva showing lumen of gut packed with gregarines. What features of gregarine structure can you identify. 4. Section of midgut of meal worm (Tenebrio) larva showing lumen of gut packed with gregarines. What features of gregarine structure can you identify. 5. Monocystis agilis. Young trophozoites inside sperm morulae from seminal vesicle of earth worm. Note sperm morulae (clusters of spermatids) with and without gregarines. At later stages in sperm differentiation the sperm nuclei look like purple hairs attached to the surface of the pink trophozoites. Note small sporadin (slug-shaped between center and 3:00 Slide Z 86.2 x 16.6 6. Note small sporadin (worm-shaped) near center. Slide 1 91.3 x 16.2 7. Monocystis agilis. Young trophozoites inside sperm morulae from seminal vesicle of earth worm. Note sperm morulae (clusters of spermatids) with and without gregarines. At later stages in sperm differentiation the sperm nuclei look like purple hairs attached to the surface of the pink trophozoites 8. Monocystis agilis. Whole mount . Older trophozoite (sporidin). This is the large purple spindle-shaped cell. slide W1, 8.2 x 112.0. 9. Monocystis agilis Whole mount slide 5, 90.4 x 12.2. Gamonts in syzygy. 10. Monocystis agilis Whole mount slide 2, 14.7 x 106. Gamontocyst with gamonts in syzygy. 11. Feel free to move this slide. 12 Monocystis agilis. Whole mount slide 4, 84.2 x 12.2. Gamontocyst showing gametes budding off of the central cytoplasmic mass. Note: Two gamontocysts are shown here. The one on the right shows gametogenesis, the one on the left contains mature spores that do not take up stain. Inside the gamontocyst on the right you can make out the two gamonts, each of which is giving rise to gametes. One of the gamonts lies obliquely above the other. 13. Section of gamonts in gamontocyst prior to start of gametogenesis. Notice large central nucleus on the section of one of the gamonts. Slide Q 88.3 x 11.8 14. Early gametogenesis. Nuclei have divided several times and have moved to the periphery of the gamonts. Slide 12 (red) 86.0 x 10.2 15. Later gametogenesis. Cleavage and release of gametes at the surface of the gamont. Slide J 85.7 x 12.6 16. Fertilization. Pronuclei stage. In most of these cells gametes have fused but the pronuclei have not. Notice that there are two nuclei in many of the cells. In some karyogamy (nuclear fusion has occurred resulting in a single larger nucleus. Slide C 84.7 x 13.6 17. Zygotes prior to formation of spore wall. Cells at this stage are sphaerical. Slide B 87.3 x 12.0 18. Zygotes after formation of spore wall - first meiotic prophase. Cells are football - shaped at this stage. In other gamontocysts there are cells that have completed the first meiotic division (two nuclei). How can you tell this stage from cells during fertilization that also have two nuclei? There are mature spores on other gamontocysts. See map beside microscope. 85.3 x 13.2 19. Meiosis I, after Meiosis I, after Meiosis II; after 3rd sporogenic division. See map beside microscope. Slide 12 (blue) 84.7 x 11.2 20. After meiosis II; after 3rd sporogenic division. See map beside microscope. Slide D 84.8 x 14.7 21. Meiosis I, after 3rd sporogenic division and mature spores. See map beside slide. 22-23 Mature spores.
